FIELD OF SPECIALIZATION: Ultra-fast Time Domain Fluorescence Spectroscopy and Microscopy, Laser and Molecular spectroscopy, Electronics and photonics, Electronic structure calculations through computation.

PROFESSIONAL SKILLS (TEACHING/RESEARCH EXPERIENCE):

 

1.             Three year teaching experience of Electronics and Modern Physics at Undergraduate classes and Laser & Molecular spectroscopy and Quantum Mechanics in Post Graduate classes in Department of Physics, Kumaon University Nainital. India and Banaras Hindu University, Varanasi, India

 

2.             Fabrication of electronic circuits like amplifier, oscillator, power supply, timer, transmitter / receiver, ms flash lamp etc. Design, maintenance and operation of spectrophotometer and spectrofluorometer for steady state and time domain measurements.

 

3.             I have studied the various primary photochemical processes viz. Photo-induced excited state proton transfer, electron transfer, charge transfer, energy transfer, solvent relaxation etc in some hydrogen bonded molecular systems.

 

4.             Basic knowledge of Fluorescence  in situ hybridization (FISH),  Fluorescence Recovery After Photo-bleaching (FRAP), Fluorescence Resonance Energy Transfer Microscopy (FRETM), Fluorescence Lifetime Imaging Microscope (FLIM), Optical Tweezers,  X-ray crystallography, NMR spectroscopy, CD, FTIR.

 

5.             Experience in maintenance and operation of various types of fluorescence spectroscopic instruments viz. Spectrophotometer (ratio recording Backman DK-A, JASCO V-550), Spectrofluorometer (JASCO-FP-777, ISA JOBIN YVON-SPEX-FL3-22), Time domain spectrophotometer Edinburg FLA 199) with short pulse generator like micro-second, nano-second and pico second range pulse through flash lamp, (H2 and N2 gas discharge lamp) / mode locked, frequency doubled Nd-YAG laser coupled with cavity dumped dye laser, Nitrogen laser coupled with BOXCAR (SRS-245), time domain confocal fluorescence Microscope, Pico-Quaunt (Nicon fluoroscence microscope coupled with TCSPC system and pico second diode laser ) and low temperature instruments like cryostat APD – E-CS-202) respectively.

 

6.             Sample purification, crystallization and preparation of plastic films viz. protic, aprotic and ion-exchange by cast method. Preparation of various glass systems doped with organic and inorganic ions and chelet formation.  DNA, protein isolation, purification and characterization.

 

7.             Data acquisition and analysis of Steady State and Time domain results through related software for fluorescence (GRAMA/386 of Galactic), Origin 6.0 and absorption data analysis as well as important Word Processing & Mathematical data analysis. Besides this different software used for theoretical computational calculations viz. Gaussian 98, Gaussian 03, Gauss view, Molden for electronic structure exploration of molecular system.

ˇ        Past Research and Achievements

I did my Post doctoral and Ph.D. with Prof. H. B. Tripathi at the Photophysics Laboratory, Department of Physics, D.S.B. Campus, Kumaon University Nainital India in the field of ultra fast time resolved fluorescence spectroscopy of intra and inter molecular hydrogen bonded molecular system under different environmental conditions with special emphasis on the of excited state proton transfer (ESPT) and its applications. Hydrogen bonding plays a very important role for existing of life in the universe. Excited state intramolecular proton transfer, is one of the most important and characteristic relaxation route in the hydrogen bonded molecular systems.  This work was carried out during the tenure of research assistantship under the DST project “Facilities for the Peco-second spectroscopy”. Then I was awarded Senior Research Fellowship by CSIR, New Delhi, India. Besides this I had also studied role of diffusion on Förster resonance energy transfer (FRET) and its application as pH sensor with the Prof. T.C. Pant, Charge transfer studies were carried out with Prof. D.D. Pant, Emeritus Professor in the Department.  Theoretical part of the research work was carried out at IIT Kanpur, India with Prof. N. Sathyamurthy. Further, I was awarded Young Scientist project by DST India and I run this project  in Molecular Biophysics Unit, Indian Institute of Science, Bangalore, under the supervision of Prof. P. Balaram in the area of “Photoinducied dynamics of a glycolytic enzyme triosephosphate isomerase (TIM) isolated from Plasmodium falciparum”.

                         Moreover, during the course of experimental investigation it was found that the spectra obtained by advance technology based spectrofluorometer JASCO FP-777 showed many anomalies in fluorescence excitation and emission spectra which could not be removed by the manufacturer. I, thus, started a comprehensive program to remove these anomalies in the spectrofluorometer and obtained a correction coefficient data for spectral measurement for this spctrofluoremeter.
